| mutation in BRCA1 or BRCA2, genetic testing may be more straightforward | |
| than in the general human population. The best-studied case is the | |
| Ashkenazi Jewish population. A recent study at Baylor College of Medicine | |
| (Houston, Texas)1 (1. Roa BR, et al. Nature Genetics. 1996;14:185-190.) | |
| showed that, among a large population (approximately 3,000 individuals) | |
| of Ashkenazi origin, 1.09% carried the BRCA1 185delAG mutation and 1.52% | |
| carried the BRCA2 6174delT mutation.2 (2. The nomenclature for these | |
| mutations is as follows: 185delAG indicates a two base-pair deletion (of | |
| AG) at nucleotide 185 in BRCA1; 6174delT is a deletion of a T at | |
| nucleotide number 6174 in BRCA2.) No one was found to carry both | |
| mutations. These two mutations are by far the most frequent mutation | |
| alleles predisposing to heritable breast cancer among the Ashkenazi | |
| population. The study found that the BRCA1 185delAG mutation accounts for | |
| approximately 19% of breast cancer cases diagnosed before age 50 in | |
| Ashkenazi women; this figure agrees with results of two other studies | |
| (16% and 20%, respectively). | |

| The finding of a high frequency of BRCA1 185delAG in the Ashkenazi Jewish | |
| population has prompted another company (in addition to OncorMed and | |
| Myriad) to offer tests for that particular mutation to Ashkenazi women | |
| from high-risk families. In April 1996, Genetics and IVF Institute | |
| (Fairfax, Virginia) announced the availability of such a test. (Since | |
| then, offerings have been broadened to include three other mutations | |
| found in relatively high frequency in the same population.) | |
